Brief presentation of the institution

The Laboratory of Biophysics and Cell Biotechnology has been from the begining focused on effects of electric field on living matter at cellular and molecular level. Main topics now: cell electroporation mechanism, non-thermal microwave (2.45 GHz) effects at cell level, cell membrane fluidity in normal and pathological states. The Laboratory is part of the 1st Preclinic Department on Functional Sciences, Carol Davila University of Medicine and Pharmacy, Bucharest, Romania. The team include biophysicists, physicians, chemists, all the members being involved both in research and educational activities (the Master Course in Medical Biophysics and Cellular Biotechnology). Master course and PhD students are involved in the research projects of the group.

Main Electroporation related topics

Related to electroporation, our group is interested in studying the changes of cell dielectric and optical properties induced by electroporation by using  dielectrophoresis and optical trapping techniques. We are also interested in lipid organization in the membrane of living cells or in cellular modelsas reflected by parameters like: membrane fluidity, generalized polarization after the application of porating electric pulses.

Main Electroporation related research techniques and equipment

- electroporators: Electro Cell B10 BetaTech (rectangular bipolar pulses, 1-1000V, 5µs-5ms), Multiporator Eppendorf (exponentially-decaying pulses, 1-1200V, 15-500µs),

- wide field fluorescence microscopes (Zeiss Axiovert 200 and AxioObserver D1) equipped with color (AxioCam ICc3) and monochrome CCD (AxioCam MRn) cameras, with Canon A640 photocamera, and with DIC and fluorescence cubes (Zeiss 00, 02, 09, 15, Chroma 11003v2),

- cell culture laboratory: CO2 incubator, laminar flow cabinet, cooled centrifuge, visible light inverted microscope, liquid N2 containers, Beckman Coulter cell counter, TKA milliQ water source,

- function generator Agilent 33250A, oscilloscopes (PCScope PCS500 and PCSU 1000 Velleman), homemade electric generators for dielectrophoresis and electroporation/electrofusion on small volume chambers,

- dielectrophoresis chambers with various geometries of electrodes,

- ThorLab optical tweezers (945nm) and home made optical tweezers (831 nm),

- RTCA DP XCell Roche with E-plate16 and CIM-Plate 16,

- software for image acquisition and processing (Zeiss AxioVision, Image ProPlus).

- spectrophotometers: FP-6300 Able Jasco with thermostated cuvette holder and Awareness 96 Plate reader,
- spectrofluorimeters: Fluorolog 3 (Horiba Jobin Yvone) T shape with thermostated cuvette holder, SPEX T shape  with thermostated cuvette holder, ... L shape,
- cFlow Microfluidics system (Cell MicroControls) with debit and temperature control unit, ITO chambers....,
- ultracentrifuge (100 000g, Beckman),
- setups for DNA and protein electrophoresis and Vilbert Lourmat gel developer.
